Objectives
THE CLASS OF PHYSICAL METHODS IN ORGANIC CHEMISTRY IS AIMED AT ACQUIRE KNOWLEDGE AND UNDERSTANDING OF THE THEORY AND APPLICATIONS OF INSTRUMENTAL TECHNIQUES USEFUL FOR QUALI-QUANTITATIVE CHARACTERIZATION OF ORGANIC COMPOUNDS AND BIOMOLECULES, THROUGH SPECTROSCOPY (IR, UV, CD, NMR) AND MASS SPECTROMETRY.
THROUGH THE APPLICATION OF THE ACQUIRED SKILLS, STUDENTS MUST BE ABLE TO SELECT AND COMBINE THE MAIN INSTRUMENTAL APPROACHES TO THE SOLUTION OF ANALYTICAL PROBLEMS IN BIOORGANIC CHEMISTRY

Prerequisites
KNOWLEDGE OF GENERAL, INORGANIC AND ORGANIC CHEMISTRY IS REQUIRED. PASSING ALL THE FIRST YEAR CLASSES, ORGANIC CHEMISTRY I, AND GENERAL BIOCHEMISTRY IS MANDATORY.

2: INFRARED SPECTROSCOPY (IR): MODEL DESCRIPTION OF THE VIBRATIONAL PROPERTIES: THE CLASSICAL AND QUANTUM HARMONIC OSCILLATOR. THE USE OF IR FOR CHEMICAL DIAGNOSTICS. APPLICATIONS. (6 HOURS)
3: ULTRAVIOLET-VISIBLE SPECTROSCOPY (UV-VIS): MOLECULAR ORBITALS AND LCAO THEORY. ELECTRONIC TRANSITIONS. BEER-LAMBERT LAW. QUANTITATIVE ASPECTS OF UV SPECTROSCOPY IN RELATION TO QUALITY CONTROL. PREDICTION OF ABSORBANCE LAMBDA-MAX THROUGH WOODWARD RULES. APPLICATIONS. (6 HOURS)
4: CIRCULAR DICHROISM (CD) SPECTROSCOPY: PRINCIPLES OF CIRCULAR DICHROISM (CD) AND POLARIMETRY. OPERATION AND USE OF A CD SPECTROPHOTOMETER. USING CD SPECTROSCOPY FOR THE DETERMINATION OF THE CONFORMATION AND CONFIGURATION OF ORGANIC MOLECULES. (2 HOURS)
5: SURFACE PLASMON RESONANCE (SPR): GENERAL PRINCIPLES, INSTRUMENTATION AND SCOPE. (2 HOURS)
6: MASS SPECTROMETRY: ION SOURCES AND ANALYZERS. NOTES ON THE PHYSICAL THEORY CONCERNING THE PRODUCTION AND THE OBSERVATION OF MOLECULAR IONS. IONIZATION TECHNIQUES. ANALYZERS. TANDEM MASS SPECTROMETRY AND APPLICATIONS. (12 HOURS)
8.NUCLEAR MAGNETIC RESONANCE (NMR): GENERAL CHARACTERISTICS AND THEORY OF CHEMICAL SHIFTS AND COUPLING OF SPIN. 1H-NMR AND 13C-NMR. TWO-DIMENSIONAL NMR SPECTROSCOPY. APPLICATIONS. (18 HOURS)
Teaching Methods
THIS CLASS (6 CFU) CONSISTS OF 48 HOURS OF LECTURES SUPPORTED BY SLIDE PRESENTATION. CLASS ATTENDANCE IS COMPULSORY. STUDENTS ARE ALLOWED TO GET THE EXAMINATION IF THEIR ATTENDANCE IS GREATER THAN 70%. CLASS ATTENDANCE IS CHECKED ONLINE
Verification of learning
THE EXAM IS COMPOSED BY A WRITING AND AN ORAL TEST. THE FIRST STEP IS THE WRITING TEST, PREPARATORY FOR THE ORAL EXAM AND CONSISTS OF ESSAY QUESTIONS ON THE TOPICS STUDIED. IT IS MADE OF THE FOLLOWING QUESTIONS:
A) IR SPECTROSCOPY (MAX 4 POINTS)
B) UV SPECTROSCOPY (MAX 4 POINTS)
C) MASS SPECTROMETRY (MAX 8 POINTS)
D) NMR SPECTROSCOPY (MAX 5 POINTS)
E) SPECTRAL DATA OF ORGANIC MOLECULES (MAX 5 POINTS)
F) SHORT ANSWERS QUESTIONS (MAX 4 POINTS)
THE WRITING TEST IS PASSED WITH A MINIMUM SCORE OF 18/30. IT LASTS 90 MINUTES AND IS DUE TO UNDERSTAND THE STUDENT COMPREHENSION OF THE TOPICS AND HIS ABILITY IN THE APPLICATION OF THEORETICAL KNOWLEDGE. THE ORAL TEST WILL BE ACCESSIBLE ONLY TO STUDENTS REACHING THE MINIMUM SCORE IN THE WRITING TEST AND CONSISTS IN QUESTIONS ON THE TOPICS STUDIED, TO EVALUATE THE STUDENTS LEVEL OF KNOWLEDGE AND COMPREHENSION, AND THEIR PRESENTATION SKILLS USING A CORRECT SCIENTIFIC LANGUAGE. THE ORAL TEST IS AS WELL SCORED ION A SCALE OF 30. THE FINAL GRADE IS THE WEIGHED MEAN (WRITING: ORAL TESTS 1:2) OF THE GRADES GAINED IN EACH TEST.
