COURSE AIM:THE COURSE AIMS TO PROVIDE THE STUDENTS WITH THE BASIC KNOWLEDGE NEEDED TO ADDRESS THE STRUCTURE DETERMINATION OF BIOLOGICAL MOLECULES THROUGH ANALYSIS OF THEIR SPECTROSCOPIC PROPERTIES.
KNOWLEDGE AND UNDERSTANDING: THE MAIN ACQUIRED KNOWLEDGE WILL BE:
• KNOWLEDGE OF THE FOUNDATIONAL PRINCIPLES THAT GOVERN BOTH THE THEORETICAL AND EXPERIMENTAL ASPECTS OF NUCLEAR MAGNETIC RESONANCE (NMR), INFRARED SPECTROSCOPY (IR), ULTRAVIOLET AND VISIBLE SPECTROSCOPY (UV-VIS) AND MASS SPECTROMETRY (MS);
• KNOWLEDGE OF THE POTENTIALITIES AND LIMITS OF APPLICATIONS OF THE DIFFERENT SPECTROSCOPIC TECHNIQUES IN CORRELATION OF THE BIOMOLECULAR SYSTEMS TO BE ANALYZED AND THE INFORMATIONTO BE OBTAINED;
•HOW TO CARRY A COMPARATIVE ANALYSIS OF THE DIFFERENT SPECTROSCOPIC TECNIQUES TO OBTAIN INFORMATION ON THE STRUCTURAL AND DYNAMIC CHARACTERIZATION OF BIOMOLECULES;
•KNOWLEDGE OF THE ESSENTIAL INSTRUMENTATIONT FOR CONDUCTING NMR, IR, UV AND MS SPECTROSCOPY EXPERIMENTS.
APPLYING KNOWLEDGE AND UNDERSTANDING: THE STUDENT WILL BE ABLE:
•TO SELECT THE SPECTROSCOPIC TECHNIQUE SUITABLE FOR THE SAMPLE INVESTIGATION ON THE BASIS OF THEIR STRUCTURAL FEATURES, SAMPLE AVAILABILITY AND TYPE OF OUTPUT REQUESTED.
•TO SET UP AND IMPLEMENT NMR, IR, UV AND MS SPECTROSCOPY EXPERIMENTS.
•TO INTERPRET EXPERIMENTAL DATA.
•TO SOLVE STRUCTURAL PROBLEMS BY SPECTROSCOPIC ANALYSIS.
MAKING JUDGMENTS :THE STUDENT WILL BE ABLE:
•TO CRITICALLY EVALUATE SCIENTIFIC ARTICLES THAT REPORT RESULTS OBTAINED USING SPECTROSCOPIC METHODS RELATED TO THE COURSE TOPICS
•TO ANALYZE A STRUCTURAL ISSUE AND CHOOSE THE SPECTROSCOPIC METHOD BEST SUITED FOR THE RESOLUTION, TAKING INTO ACCOUNT THE CHARACTERISTICS OF THE SAMPLE TO BE ANALYZED
•TO ANALYZE DATA RESULTING FROM THE SPECTROSCOPIC METHODS INDICATED ABOVE.
COMMUNICATION SKILLS :THE STUDENT WILL BE ABLE:
•TO EXPLAIN THE EXPERIMENTAL RESULTS FROM THE COMPLEMENTARY USE OF VARIOUS SPECTROSCOPIC METHODS WITH APPROPRIATE SCIENTIFIC LANGUAGE
LEARNING SKILL:THE STUDENT WILL BE ABLE:
•TO USE THE TEACHING MATERIAL PROVIDED BY THE TEACHER INDEPENDENTLY AND CRITICALLY.
•TO FIND AND INTERPRET ADDITIONAL INFORMATION RELATED TO THE TOPICS COVERED BY THE COURSE.
Prerequisites
A BASIC KNOWLEDGE OF INORGANIC CHEMISTRY, ORGANIC CHEMISTRY, BIOCHEMISTRY, AND GENERAL PHYSICS
Contents
THE TOPICS ADDRESSED IN THE COURSE ARE:
GENERAL PRINCIPLES OF SPECTROSCOPY. NUCLEAR MAGNETIC RESONANCE., 1H-NMR AND 13C-NMR AND MULTIPULSE MULTIDIMENSIONAL NMR TECHNIQUES (12H LECTURE, 3H LABORATORY, 7H EXERCITATION). MASS SPECTROMETRY: INTRODUCTION, IONISATION PROCESSES, FRAGMENTATION PROCESSES (8H LECTURE, 3H LABORATORY, 5H EXERCITATION), APPLICATIONS. ULTRAVIOLET AND VISIBLE SPECTROSCOPY: MOLECULAR ORBITAL AND ELECTRONIC TRANSITIONS, CHROMOPHORES,APPLICATIONS (6H LECTURE, 3H LABORATORY, 2H EXERCITATION). INFRARED SPECTROSCOPY, MOLECULAR VIBRATIONS ,STRUCTURAL INFORMATIONS IN MACROMOLECULES WITH VIBRATIONAL SPECTROSCOPY (6H LECTURE, 3H LABORATORY, 3H EXERCITATION).
Teaching Methods
COURSE WORKLOAD: 56H. THE COURSE WILL BE TAUGHT IN A COMBINATION OF LECTURES (32H, 4CFU), CLASSROOM EXERCISES AND PRACTICAL SESSIONS (24H, 2 CFU). THE INTERACTIVE CLASSROOM EXERCISES (12 HOURS) CONCERN THE ANALYSIS OF SPECTRAL PROBLEMS RELATED TO STRUCTURE DETERMINATION OF ORGANIC MOLECULE; IN THE LABORATORY SESSION (12 HOURS) THE STUDENTS CARRY OUT SPECTROSCOPY EXPERIMENTS WITH SUPERVISION.
Verification of learning
THE FINAL EVALUATION WILL BE PERFORMED BY AN ORAL INTERVIEW. THE EXAM CONSISTS: OF THE RESOLUTION OF THE STRUCTURE OF BIOMOLECULES BY ANALYSIS OF NMR, IR, UV AND MASSA SPECTRA AND AND STARTING FROM WHAT EMERGED DURING THE DISCUSSION, OTHER ISSUES ARE ADDRESSED RELATED TO THE COURSE TOPICS. THE FINAL ASSESSMENT IS EVALUATED IN THIRTY AND TAKES INTO ACCOUNT THE DEGREE OF UNDERSTANDING OF THE STUDENT, THE EXACTNESS OF THE TERMS USE, CRITICAL THINKING AND ABILITY TO CREATE AND UNDERSTAND CONNECTIONS TO OTHER ISSUES ADDRESSED DURING THE DISCUSSION.
